RESUMO

Management of thorny skate (Amblyraja radiata) in the Northwest Atlantic has posed a conservation dilemma for several decades due to the species' lack of response to strong conservation efforts in the US Gulf of Maine and the Canadian Scotian Shelf, confusion over the relationship between two reproductive size morphs of differing life histories that are sympatric in the Northwest Atlantic, and conflicting data on regional population connectivity throughout the species' broader range. To better assess potential A. radiata regional population differentiation and genetic links to life-history variation, we analysed complete mitochondrial genome sequences from 527 specimens collected across the species' North Atlantic geographic range, with particular emphasis on the Northwest Atlantic region. A high level of genetic diversity was evident across the North Atlantic, but significant genetic differentiation was identified between specimens inhabiting the Northwest (Gulf of Maine and Newfoundland) and Northeast (Greenland, Iceland, North Sea, and Arctic Circle) Atlantic. In the Northwest Atlantic, significant differentiation between the Gulf of Maine and Newfoundland regions was revealed; however, the overall level of differentiation was very low. No genetic difference was identified between the large and small reproductive morphs. The results of this study advance our understanding of A. radiata population structure in the North Atlantic but do not resolve all the questions confounding our understanding of the species' biology and evolutionary history.

The gill slits of fishes develop from an iterative series of pharyngeal endodermal pouches that contact and fuse with surface ectoderm on either side of the embryonic head. We find in the skate (Leucoraja erinacea) that all gill slits form via a stereotypical sequence of epithelial interactions: 1) endodermal pouches approach overlying surface ectoderm, with 2) focal degradation of ectodermal basement membranes preceding endoderm-ectoderm contact; 3) endodermal pouches contact and intercalate with overlying surface ectoderm, and finally 4) perforation of a gill slit occurs by epithelial remodelling, without programmed cell death, at the site of endoderm-ectoderm intercalation. Skate embryos express Fgf8 and Fgf3 within developing pharyngeal epithelia during gill slit formation. When we inhibit Fgf signalling by treating skate embryos with the Fgf receptor inhibitor SU5402 we find that endodermal pouch formation, basement membrane degradation and endodermal-ectodermal intercalation are unaffected, but that epithelial remodelling and gill slit perforation fail to occur. These findings point to a role for Fgf signalling in epithelial remodelling during gill slit formation in the skate and, more broadly, to an ancestral role for Fgf signalling during pharyngeal pouch epithelial morphogenesis in vertebrate embryos.

The retinas of the vast majority of vertebrate species are termed "duplex," that is, they contain both rod and cone photoreceptor neurons in different ratios. The retina of little skate (Leucoraja erinacea) is a rarity among vertebrates because it contains only a single photoreceptor cell type and is thus "simplex." This unique retina provides us with an important comparative model and an exciting opportunity to study retinal circuitry within the context of a visual system with a single photoreceptor cell type. What is perhaps even more intriguing is the fact that the Leucoraja retina is able use that single photoreceptor cell type to function under both scotopic and photopic ranges of illumination. Although some ultrastructural characteristics of skate photoreceptors have been examined previously, leading to a general description of them as "rods" largely based on outer segment (OS) morphology and rhodopsin expression, a detailed study of the fine anatomy of the entire cell and its synaptic connectivity is still lacking. To address this gap in knowledge, we performed serial block-face electron microscopy imaging and examined the structure of skate photoreceptors and their postsynaptic partners. We find that skate photoreceptors exhibit unusual ultrastructural characteristics that are either common to rods or cones in other vertebrates (e.g., outer segment architecture, synaptic ribbon number, terminal extensions), or are somewhere in between those of a typical vertebrate rod or cone (e.g., number of invaginating contacts, clustering of multiple ribbons over a single synaptic invagination). We suggest that some of the ultrastructural characteristics we observe may play a role in the ability of the skate retina to function across scotopic and photopic ranges of illumination. Our findings have the potential to reveal as yet undescribed principles of vertebrate retinal design.

Prior research has demonstrated that male adults and youth engaged in skateboarding at community skateparks achieve heart rates that meet or exceed recommendations for exercise by the CDC. However, these studies do not adequately evaluate other non-traditional or ethnically diverse users who may differ in how they utilize the skatepark and in their cardiovascular response. The purpose of this experiment was to measure heart rate response and locomotor movement in three lesser studied groups that frequently utilize community skateparks, and to compare these results with those reported previously in male adult and youth skateboarders. Fifty-six skatepark users were analyzed, including thirty female and BIPOC skateboarders, and twenty-six non-skateboard users. All participants were instrumented with a HR monitor with GPS capability and asked to engage in their preferred activity with no duration specified. Average heart rate and time spent at high and moderate levels of heart rate intensity were not statistically different among the groups studied here, nor were they different from those reported previously for male adult and youth skateboarders. Distances traveled, average, and peak velocities were also not statistically different among the groups studied here, but all were significantly lower than values previously reported for adult male skateboarders. While some differences in distances traveled and velocity were noted, all groups met or exceeded CDC guidelines for cardiovascular fitness. These data suggest that skateparks can help a community achieve health outcomes, particularly among diverse users.

Members of the family Polyomaviridae have a circular double-stranded DNA genome that have been identified in various hosts ranging from mammals to arachnids. Here we report the identification and analysis of a complete genome sequence of a novel polyomavirus, Raja clavata polyomavirus (RcPyV1), from a cartilaginous fish, the thornback skate (Raja clavata). The genome sequence was determined using a metagenomics approach with an aim to provide baseline viral data in cartilaginous fish in different ecosystems. The RcPyV1 genome (4,195 nucleotides) had typical organization of polyomavirus, including early antigens (small T; Large T) encoded on one strand and late viral proteins (VP1; VP2) on the complementary strand. Maximum-likelihood phylogenetic analysis of the large T-antigen revealed that RcPyV1 clusters with a polyomavirus obtained from another cartilaginous fish, the guitarfish polyomavirus 1 (GfPyV1). These two share ~ 56% pairwise identity in LT and VP1 protein sequences. These analyses support the hypothesis that cartilaginous fishes have a specific lineage of polyomaviruses.

Collagen peptides are widely employed as therapeutic materials due to their numerous beneficial properties, including for the following uses: antiaging, antioxidant applications, antibacterial applications, wound healing, tissue engineering, medication delivery, and cosmetics. Although collagen peptides are useful in these applications, to our knowledge, few published studies have been undertaken on their repeated-dose toxicity. We evaluated the possible subchronic toxicity of a collagen peptide derived from skate (Raja kenojei) skin (CPSS) in Sprague-Dawley rats by administering repeated oral doses over 90 days. Rats of both sexes were assigned randomly to one of four experimental groups, respectively receiving 0, 500, 1000, or 2000 mg/kg/day of CPSS. At all doses tested, repeated oral CPSS administration had no treatment-related adverse effects in terms of clinical signs, body weight, food consumption, detailed clinical observation, sensory reactivity, functional assessment, urinalysis, ophthalmic examination, gross pathology, hematology, serum biochemistry, hormone analysis, organ weight, and histopathology. Even though there were some alterations in hematologic parameters, serum biochemistry parameters, organ weight, and histopathological findings, these did not follow a dose-response pattern and were within historical limits for control rats. The oral no-observed-adverse-effect level (NOAEL) of the CPSS was 2000 mg/kg/day for both male and female rats in the applied experimental circumstances, and no target organs were identified.

Despite a high species diversity, skates (Rajiformes) exhibit remarkably conservative morphology and ecology. Limited trait variations occur within and between species, and cryptic species have been reported among sister and non-sister taxa, suggesting that species complexes may be subject to stabilising selection. Three sibling species are currently recognised in the Raja miraletus complex: (i) R. miraletus occurring along the Portuguese and Mediterranean coasts, (ii) R. parva in the Central-Eastern Atlantic off West Africa and (iii) R. ocellifera in the Western Indian Ocean off South Africa. In the present study, the genetic variation at mitochondrial and nuclear markers was estimated in the species complex by analysing 323 individuals sampled across most of its geographical distribution area to test the hypothesis that restricted gene flow and genetic divergence within species reflect known climate and bio-oceanographic discontinuities. Our results support previous morphological studies and confirm the known taxonomic boundaries of the three recognised species. In addition, we identified multiple weakly differentiated clades in the Northeastern Atlantic Ocean and Mediterranean, at least two additional cryptic taxa off Senegal and Angola, a pronounced differentiation of ancient South African clades. The hidden genetic structure presented here may represent a valuable support to species' conservation action plans.

O objetivo da pesquisa foi analisar o perfil da produção do conhecimento sobre skate em artigos científicos em português indexados em periódicos brasileiros. As buscas ocorreram nas bases de dados científicos BASE, BVS, Portal de periódicos CAPES, DOAJ, REDALYC e SciELO. Ao todo foram incluídos 64 artigos científicos e a análise abordou a distribuição de artigos anualmente, os tipos de metodologia, os artigos mais citados, os periódicos científicos com mais publicações sobre o skate, os autores e filiações, e os enfoques temáticos das pesquisas. Observou-se que os estudos sobre skate foram publicados entre 2008 e 2022, com prevalência de pesquisas qualitativas (86%), especialmente as relacionadas com aspectos sociais, culturais e históricos (73%), publicadas em periódicos da área da Educação Física e da Antropologia/Arqueologia, e com autores vinculados predominantemente às instituições das regiões Sudeste e Sul do Brasil.


The aim of the research was to analyze the profile of the production of knowledge about skateboarding in scientific articles in Portuguese indexed in Brazilian journals. The searches were performed on the BASE, BVS, CAPES, DOAJ, REDALYC and SciELO databases. In total, 64 scientific articles were selected. The analysis addressed the distribution of articles annually, the types of methodology, the most cited articles, the scientific journals with the most publications, the authors and affiliations, and the thematic approaches of the researchers. The skateboarding studies were published between 2008 and 2022, with a prevalence of qualitative research (86%), primarily related to social, cultural and historical aspects (73%). The articles were mostly published in journals in the field of Physical Education and Anthropology/Archaeology and with authors linked to institutions in Brazil's Southeast and South regions of Brazil.

Roller speed skating is a discipline in which muscle fatigue plays an important role in athletes; in this work, we wanted to evaluate whether a methodological approach based on the energy required to maintain an isometric muscle contraction for one minute, indexed on the MVIC (maximum voluntary isometric contraction), i.e. % RMS/MVIC can give results similar to the frequency decay analysis/time in terms of usability for athlete training and injury prevention. Right and left gluteus maximus and vastus lateralis muscles (involved in the propulsive phase of skating ) were examined separately by surface electromyography in three competitive athletes in short-track speed skating on asphalt. The results showed an asymmetry between the right (less resistant) and left (more resistant) lower limb, in all three athletes, from the point of view of fatigue, in both investigated muscles. Furthermore, a trend in terms of fatigue resistance was found that was directly proportional to skill in both muscles studied. This can be of help in better planning the training of street speed skaters (which although similar to the discipline on ice is not completely superimposable) with the dual purpose of improving their performance and preventing injuries, often linked to the degree of right-left muscle asymmetry.

The gill skeleton of cartilaginous fishes (sharks, skates, rays, and holocephalans) exhibits a striking anterior-posterior polarity, with a series of fine appendages called branchial rays projecting from the posterior margin of the gill arch cartilages. We previously demonstrated in the skate (Leucoraja erinacea) that branchial rays derive from a posterior domain of pharyngeal arch mesenchyme that is responsive to Sonic hedgehog (Shh) signaling from a distal gill arch epithelial ridge (GAER) signaling centre. However, how branchial ray progenitors are specified exclusively within posterior gill arch mesenchyme is not known. Here, we show that genes encoding several Wnt ligands are expressed in the ectoderm immediately adjacent to the skate GAER, and that these Wnt signals are transduced largely in the anterior arch environment. Using pharmacological manipulation, we show that inhibition of Wnt signalling results in an anterior expansion of Shh signal transduction in developing skate gill arches, and in the formation of ectopic anterior branchial ray cartilages. Our findings demonstrate that ectodermal Wnt signalling contributes to gill arch skeletal polarity in skate by restricting Shh signal transduction and chondrogenesis to the posterior arch environment and highlights the importance of signalling interactions at embryonic tissue boundaries for cell fate determination in vertebrate pharyngeal arches.

Estimating the demographic parameters of contemporary populations is essential to the success of elasmobranch conservation programmes, and to understanding their recent evolutionary history. For benthic elasmobranchs such as skates, traditional fisheries-independent approaches are often unsuitable as the data may be subject to various sources of bias, whilst low recapture rates can render mark-recapture programmes ineffectual. Close-kin mark-recapture (CKMR), a novel demographic modelling approach based on the genetic identification of close relatives within a sample, represents a promising alternative approach as it does not require physical recaptures. We evaluated the suitability of CKMR as a demographic modelling tool for the critically endangered blue skate (Dipturus batis) in the Celtic Sea using samples collected during fisheries-dependent trammel-net surveys that ran from 2011 to 2017. We identified three full-sibling and 16 half-sibling pairs among 662 skates, which were genotyped across 6291 genome-wide single nucleotide polymorphisms, 15 of which were cross-cohort half-sibling pairs that were included in a CKMR model. Despite limitations owing to a lack of validated life-history trait parameters for the species, we produced the first estimates of adult breeding abundance, population growth rate, and annual adult survival rate for D. batis in the Celtic Sea. The results were compared to estimates of genetic diversity, effective population size (N e ), and to catch per unit effort estimates from the trammel-net survey. Although each method was characterized by wide uncertainty bounds, together they suggested a stable population size across the time-series. Recommendations for the implementation of CKMR as a conservation tool for data-limited elasmobranchs are discussed. In addition, the spatio-temporal distribution of the 19 sibling pairs revealed a pattern of site fidelity in D. batis, and supported field observations suggesting an area of critical habitat that could qualify for protection might occur near the Isles of Scilly.

Onchobothrium malakhovin. sp. was found in the spiral valve of the softnose skate Bathyraja (Arctoraja) sexoculata off the Simushir Island (Kuril Islands, Russia). The new species has bothridia with three loculi and no additional suckers on bothridia, single-toothed hooks unconnected by their bases, no spines at the bases of the hooks, dense matrix around the hook bases shaped as an unpaired butterfly wing, and a short and wide ovary. Onchobothrium malakhovin. sp. differs from O. antarcticum and O. magnum in having a smaller total length, cirrus sac and ovary, smaller testes and eggs. Additionally, the new species differs from O. antarcticum by the absence of a vaginal sphincter and shorter bothridia; differs from O. magnum in having fewer proglottids and smaller vitelline follicles. It differs from O. farmeri, O. convolutum, and O. pseudouncinatum, by the absence of a small spine at the base of the hooks and the absence of accessory suckers on bothridia; from O. pseudouncinatum, additionally, by unconnected hooks; from O. schizacanthium, by the number of testes and by the presence of a postvaginal group of testes. Onchobothrium malakhovin. sp. was placed among other members of the Onchoproteocephalidea with a high support based on the sequence data for the D1-D3 region of the 28S rDNA and cox1 gene. The phylogenetic position of the genus Onchobothrium sensu lato remains ambiguous. We suggest that Onchobothrium sensu lato is a complex genus containing at least two morphologically different groups of species. Onchobothrium farmer, O. convolutum, O. schizacanthium, and O. pseudouncinatum, for which there are no molecular genetic data, are considerably different morphologically from O. malakhovin. sp., O. antarcticum, and O. magnum. A new genus might have to be established for the latter three species after the accumulation of genetic data.

This study aims to determine the age and growth characteristics of Dipturus oxyrinchus living in the Northeastern Mediterranean Sea and to present data that can provide a comparison with previous studies on the same subject. A total of 255 long-nose skates at a total length of 12.2-93.5 cm and weight of 8.34-3828 g were collected as non-target species from a commercial fishing boat. The male-female ratio was determined as 1:1.27. Using the von Bertalanffy equation and the Gompertz or logistic growth models, the growth parameters of Dipturus oxyrinchus were estimated as L∞ = 154.0, K = 0.064, t0 = -1.622; L∞ = 104.0, K = 0.35, I = 4.99; L∞ = 128.40, K = 0.19, I = 4.39 for all individuals, respectively. Maximum absolute growth was calculated as 9.33 cm at 5-6 years of age. Maximum relative growth at 1-2 years of age was estimated as 36.39%. Both absolute and relative growth were minimal in the 11-12 age group. The highest condition factor value was estimated as 0.416 in the 8-year-old group. As a result, the growth data of long-nose skates were obtained for the first time in the Northeastern Mediterranean Sea.

The little skate Leucoraja erinacea, a cartilaginous fish, displays pelvic fin driven walking-like behavior using genetic programs and neuronal subtypes similar to those of land vertebrates. However, mechanistic studies on little skate motor circuit development have been limited, due to a lack of high-quality reference genome. Here, we generated an assembly of the little skate genome, with precise gene annotation and structures, which allowed post-genome analysis of spinal motor neurons (MNs) essential for locomotion. Through interspecies comparison of mouse, skate and chicken MN transcriptomes, shared and divergent gene expression profiles were identified. Comparison of accessible chromatin regions between mouse and skate MNs predicted shared transcription factor (TF) motifs with divergent ones, which could be used for achieving differential regulation of MN-expressed genes. A greater number of TF motif predictions were observed in MN-expressed genes in mouse than in little skate. These findings suggest conserved and divergent molecular mechanisms controlling MN development of vertebrates during evolution, which might contribute to intricate gene regulatory networks in the emergence of a more sophisticated motor system in tetrapods.

The pseudobranch is a gill-like epithelial elaboration that sits behind the jaw of most fishes. This structure was classically regarded as a vestige of the ancestral gill arch-like condition of the gnathostome jaw. However, more recently, hypotheses of jaw evolution by transformation of a gill arch have been challenged, and the pseudobranch has alternatively been considered a specialised derivative of the second (hyoid) pharyngeal arch. Here, we demonstrate in the skate (Leucoraja erinacea) that the pseudobranch does, in fact, derive from the mandibular arch, and that it shares gene expression features and cell types with gills. We also show that the skate mandibular arch pseudobranch is supported by a spiracular cartilage that is patterned by a shh-expressing epithelial signalling centre. This closely parallels the condition seen in the gill arches, where cartilaginous appendages called branchial rays, which support the respiratory lamellae of the gills, are patterned by a shh-expressing gill arch epithelial ridge. Together with similar discoveries in zebrafish, our findings support serial homology of the pseudobranch and gills, and an ancestral origin of gill arch-like anatomical features from the gnathostome mandibular arch.

We describe the complete mitochondrial genomes of the flapper skate Dipturus intermedius (Parnell 1837) and the longnose skate Dipturus oxyrinchus (Linnaeus 1758), which have been obtained by Sanger sequencing. We report the length of the sequences to be 16,906 and 16,911 bp, respectively. The length and structure of gene regions, containing 13 protein-coding regions, 22 tRNA genes, two rRNA genes, and two non-coding areas, resemble those of related skate species. Despite D. intermedius being considered a cryptic species with D. batis, the full mitogenomes confirm that D. intermedius and D. oxyrinchus are more genetically similar. In comparison to other Dipturus species, D. intermedius is missing a whole codon in its cytochrome oxidase subunit 2 gene. These mitogenomes will be a useful resource furthering investigation of the population genetic differences and evolutionary history of skate species.

The vast majority of extant vertebrate diversity lies within the bony and cartilaginous fish lineages of jawed vertebrates. There is a long history of elegant experimental investigation of development in bony vertebrate model systems (e.g., mouse, chick, frog and zebrafish). However, studies on the development of cartilaginous fishes (sharks, skates and rays) have, until recently, been largely descriptive, owing to the challenges of embryonic manipulation and culture in this group. This, in turn, has hindered understanding of the evolution of developmental mechanisms within cartilaginous fishes and, more broadly, within jawed vertebrates. The little skate (Leucoraja erinacea) is an oviparous cartilaginous fish and has emerged as a powerful and experimentally tractable developmental model system. Here, we discuss the collection, husbandry and management of little skate brood stock and eggs, and we present an overview of key stages of skate embryonic development. We also discuss methods for the manipulation and culture of skate embryos and illustrate the range of tools and approaches available for studying this system. Finally, we summarize a selection of recent studies on skate development that highlight the utility of this system for inferring ancestral anatomical and developmental conditions for jawed vertebrates, as well as unique aspects of cartilaginous fish biology.

This study examined the micro-pacing strategies during a distance freestyle cross-country (XC) skiing competition. Nine female and 10 male highly trained XC skiers wore a GNSS device during a FIS-sanctioned race. The course was ~4900 m; women completed two-laps; men completed three-laps. The course was divided into uphill (S1, S3, S5, S7), downhill (S2, S4, S6, S8), and flat (S9) sections for analyses. Statistical parametric mapping was used to determine the course positions (clusters) where total race time or section time was significantly associated with instantaneous skiing speed. Total race time was associated with instantaneous skiing speed during a cluster in S1 on lap 2 for both sexes (t ≥ 5.899, p ≤ 0.008). The two longest uphill sections (S1; S5) and the flat section (S9) contained clusters where section times were related to instantaneous skiing speed for both sexes (p < 0.05). The fastest woman gained 6.9 s on the slowest woman during a cluster in S1 on lap 1 and 7.3 s during a cluster in S9 on lap 1. The fastest man gained 51.7 s on the slowest man over all clusters in S5 over the 3 laps combined. Compared to skiers with longer total race times, skiers with shorter race times skied with faster instantaneous speeds in some clusters of the uphill sections, as well as on the flat section of the course. This study also identified different relative micro-pacing strategies for women and men during freestyle distance XC skiing races. Finally, statistical parametric mapping analyses can help to identify individual strengths and weaknesses for guiding training programs and optimise competition pacing strategies.

The blue skate (Dipturus batis) has a patchy distribution across the North-East Atlantic Ocean, largely restricted to occidental seas around the British Isles following fisheries-induced population declines and extirpations. The viability of remnant populations remains uncertain and could be impacted by continued fishing and by-catch pressure, and the projected impacts of climate change. We genotyped 503 samples of D. batis, obtained opportunistically from the widest available geographic range, across 6 350 single nucleotide polymorphisms (SNPs) using a reduced-representation sequencing approach. Genotypes were used to assess the species' contemporary population structure, estimate effective population sizes and identify putative signals of selection in relation to environmental variables using a seascape genomics approach. We identified genetic discontinuities between inshore (British Isles) and offshore (Rockall and Faroe Island) populations, with differentiation most pronounced across the deep waters of the Rockall Trough. Effective population sizes were largest in the Celtic Sea and Rockall, but low enough to be of potential conservation concern among Scottish and Faroese sites. Among the 21 candidate SNPs under positive selection was one significantly correlated with environmental variables predicted to be affected by climate change, including bottom temperature, salinity and pH. The paucity of well-annotated elasmobranch genomes precluded us from identifying a putative function for this SNP. Nevertheless, our findings suggest that climate change could inflict a strong selective force upon remnant populations of D. batis, further constraining its already-restricted habitat. Furthermore, the results provide fundamental insights on the distribution, behaviour and evolutionary biology of D. batis in the North-East Atlantic that will be useful for the establishment of conservation actions for this and other critically endangered elasmobranchs.

The mottled skate Beringraja pulchra is a coastal fish caught by gillnet and bottom trawl fisheries in Hokkaido, Japan. We recently found a parasitic copepod identical to Pseudocharopinus markewitschi (Gusev, 1951) on this commercially important fish. A total of 109 host individuals caught off the coast of northern Hokkaido, Japan, were examined for adult females of this copepod, and 67 cases (61.5%) of infection comprised with 31 cases (67.4%) of 46 males and 36 cases (57.1%) of 63 females were found. The parasite specimens (n = 229) were recovered mainly from the dorsal side of the pelvic fin (n = 168, 73.4%), followed by the claspers (n = 32, 14.0%). The mottled skate is thought to spend most of its lifetime lying in sand or crawling on the seafloor. Therefore, the biased infection to the dorsal side of the pelvic fin may imply that site selection favors less susceptible for disturbances resulting from such host-specific behavior.
